Enhanced Humoral Immune Response by High Density TLR Agonist Presentation on Hyperbranched Polymers

چکیده
Prophylactic vaccines often exploit adjuvants to drive potent and directed immune responses. Small-molecule toll-like receptor (TLR) agonists are promising adjuvant candidates; however, the poor pharmacokinetics rapid distribution of these relatively hydrophilic small molecules results in systemic side effects that have limited their usage topical application. In this study, TLR7/8 covalently attached highly dense end-groups hyperbranched polymers synthesized by controlled radical polymerization techniques. Subcutaneous administration a prolonged type I interferon response. When co-administered model subunit vaccine, with ovalbumin or HIV envelope glycoprotein gp120, polymeric potentiate an enhanced immunoglobulin response near-immediate type-switching IgG2c, suggesting they promote strong T helper cell 1 is desirable for treatment intracellular pathogens such as mycobacterial viral infections. Controlling TLR through conjugation polymers, therefore, enables development durable high-quality humoral immunity.

PRODUCTION AND CHARACTERIZATION OF HUMORAL IMMUNE RESPONSE AGAINST MUSTARD GAS
One of the promising aspects of the immunological research on chemical war gas is to investigate the immunogenicity of some hazardous compounds such as mustard gas. Mustard gas is categorized as a "hapten" based on its physical and chemical properties. Haptenic chemicals which do not possess immunogenicity could be immunogenic experimentally when conjugated with a suitable protein carrier...
متن کاملHumoral immune response to Diphtheria and Tetanus toxoids by intranasal administration
The immunogenicity of ten different formulations of intranasal diphtheria and tetanus vaccines which containing different absorption enhancers, adjuvants and other excipients were determined in guinea pigs by the serum neutralization (SN) method. From these ten formulations, it was selected four formulations which gave significant immunogenicity in guinea pigs. In order to design the "final f...

This experiment was conducted for evaluating the humoral immune responses induced by Poly Lactide-co-Glycolide Acid (PLGA) microspheres coupled inactivated Newcastle Disease Virus (NDV) vaccine in comparison to an ‘in-house’ prepared inactivated and a live commercial vaccine. PLG microparticles containing inactivated NDV were prepared by a double emulsion technique based on solvent evaporation ...
متن کاملDendritic nanocarriers based on hyperbranched polymers.
Hyperbranched polymers are obtained through one-step polymerization reactions and exhibit properties that are very similar to those of perfect dendrimer analogues. Therefore, hyperbranched polymers are a suitable alternative for perfect dendrimers as building blocks for dendritic nanocarrier systems. With regard to using soluble hyperbranched polymers as carrier systems, their flexible chains a...
متن کاملHumoral Immune Response to AAV
Adeno-associated virus (AAV) is a member of the family Parvoviridae that has been widely used as a vector for gene therapy because of its safety profile, its ability to transduce both dividing and non-dividing cells, and its low immunogenicity. AAV has been detected in many different tissues of several animal species but has not been associated with any disease. As a result of natural infection...
